Pyro-sequencing is a method of dienei sequencing(dienei sequencing)based on the ” Sequencing by synthesis ” principle , sequencing is performed by detecting nucleotides incorporated by dienei polymerase. Pyro-sequencing relies on light detection based on chain reactions when pyrophosphate is released. Therefore, the name is sequenced into pie. ” Sequencing by synthesis ” involves taking a single strand of the gene to be sequenced and then enzymatically synthesizing its complementary strand. This method is based on detecting the activity of polymerase (polymerase synthase) in combination with other chemiluminescent enzymes. Basically , this method allows you to synthesize complementary strands along one base pair at a time and detect which bases are actually added at each step , sequencing the bases of a single strand. Nucleotides are sequentially added and removed from the reaction.
